JUBBONTI®

Classified among orthopedic injections for osteoporosis and bone-loss management, JUBBONTI® is Sandoz's denosumab-bbdz 60mg/mL prefilled syringe, approved in March 2024 as the first FDA-approved biosimilar — and the first interchangeable biosimilar — to PROLIA®. It shares the same RANKL-inhibitor mechanism, dosing schedule, and approved indications as the reference product, with biosimilar acquisition cost as the central clinical-economic argument for adoption. Why Practitioners Choose JUBBONTI®

JUBBONTI® received FDA approval as an interchangeable biosimilar to PROLIA®. Subject to applicable state laws, this designation may allow pharmacy-level substitution without prior approval from the prescriber.[1] At the time of approval, no other denosumab biosimilar had received interchangeable status, making this an important consideration for practices and payers assessing biosimilar adoption. JUBBONTI® also delivers the same molecule class, the same 60mg subcutaneous dose every six months, and the same approved indications as PROLIA®. For practices managing a high volume of patients with osteoporosis on long-term denosumab therapy, the biosimilar pathway offers a way to reduce per-dose acquisition costs without altering the treatment protocol.

What Is JUBBONTI® Used For?

JUBBONTI® is approved for the full set of PROLIA® indications. [1]
  • Postmenopausal osteoporosis at high fracture risk: The primary indication and the largest patient population for denosumab therapy. Postmenopausal bone loss accelerates with estrogen decline, and patients at high fracture risk — defined by low BMD, prior fracture history, or clinical risk factors — represent the core prescribing context for this product.
  • Men with osteoporosis at high fracture risk: Osteoporosis in men is often underrecognized and undertreated despite its contribution to fracture risk. JUBBONTI® carries the same indication as PROLIA® for this population, extending its relevance beyond postmenopausal women.
  • Glucocorticoid-induced osteoporosis: Long-term systemic corticosteroid therapy can reduce bone formation and increase bone resorption, raising fracture risk even in younger patients who may not otherwise meet typical osteoporosis criteria. This indication is particularly relevant in settings such as rheumatology, pulmonology, and transplant medicine, where chronic glucocorticoid use is common.
  • Bone loss associated with androgen deprivation therapy (ADT) in men with prostate cancer and bone loss associated with adjuvant aromatase inhibitor (AI) therapy in women with breast cancer: Sex-hormone suppression accelerates bone resorption. Oncology-adjacent practices and those managing patients on long-term hormonal cancer therapy will encounter both populations. JUBBONTI® supports bone mass preservation in these patients using the same dosing schedule as the primary osteoporosis indications.
Denosumab-bbdz is a human IgG2 monoclonal antibody that binds to RANKL and blocks its interaction with the RANK receptor. By disrupting this signaling pathway, it reduces osteoclast formation, activity, and survival, limiting bone resorption and helping preserve bone mass across its approved indications.

JUBBONTI® vs PROLIA®

This is the comparison that practitioners evaluating denosumab biosimilars are actually asking for. Both JUBBONTI® and PROLIA® (denosumab) are 60mg/mL subcutaneous injections administered every six months, using the same RANKL inhibitor pathway and carrying the same approved indications. The distinction is product origin and regulatory designation. PROLIA® is the Amgen reference product. JUBBONTI® is denosumab-bbdz, manufactured by Sandoz, with biosimilarity established through analytical, non-clinical, and clinical comparability studies demonstrating no clinically meaningful differences from the reference product. [1] In practical terms, "no clinically meaningful differences" means a practice switching from PROLIA® to JUBBONTI® should not need to modify the treatment protocol, monitoring schedule, or patient counseling — the clinical pathway remains the same. The interchangeable designation also has practical implications. Where permitted by state law, a pharmacist may substitute JUBBONTI® for PROLIA® without first contacting the prescriber. This may simplify biosimilar implementation for practices with in-house dispensing arrangements or established pharmacy partnerships. For payers and formulary managers, interchangeability provides a defined pathway for pharmacy-level substitution when clinically and legally appropriate, without requiring a new prescription for each eligible patient. Practitioners should also be clear about the distinction between JUBBONTI® and WYOST®. JUBBONTI® is the 60mg biosimilar to PROLIA®, indicated for osteoporosis and bone-loss indications. WYOST® is Sandoz's 120mg biosimilar to XGEVA®, used for prevention of skeletal-related events in oncology patients. The dose strengths, reference products, and indications differ — and this is a frequent source of confusion in clinical practice.

JUBBONTI® Safety Profile

JUBBONTI® carries the same Boxed Warning as PROLIA® for severe hypocalcemia in patients with advanced chronic kidney disease, including those on dialysis. Pre-existing hypocalcemia must be corrected before initiating therapy, and calcium levels should be monitored closely in at-risk patients. [1] Common adverse reactions include back pain, pain in extremities, musculoskeletal pain, hypercholesterolemia, and cystitis. Key risks shared with the denosumab class include osteonecrosis of the jaw, atypical femoral fracture, serious infections including skin and soft tissue infections, and dermatologic reactions. Discontinuation should be planned carefully. Stopping JUBBONTI® can lead to a rapid rebound in bone turnover and increase the risk of multiple vertebral fractures. If denosumab therapy is discontinued, patients should be transitioned to another antiresorptive treatment according to appropriate clinical guidance.[1] Contraindications include hypocalcemia, pregnancy, and known hypersensitivity to denosumab. Adequate calcium and vitamin D supplementation should be provided throughout therapy unless clinically contraindicated. [1]

About JUBBONTI® (Sandoz)

JUBBONTI® is manufactured by Sandoz, which became an independent biosimilars company following its spin-off from Novartis in October 2023. The FDA approved both JUBBONTI® and WYOST® in March 2024 as the first interchangeable biosimilars to PROLIA® and XGEVA® respectively — a significant regulatory milestone for the denosumab biosimilar landscape. [1] JUBBONTI® is supplied as a 60mg/mL single-dose prefilled syringe, matching the presentation of the PROLIA® reference product and supporting the same twice-yearly, clinic-administered treatment schedule.

JUBBONTI® Administration & Storage

  • Administer 60mg subcutaneously every 6 months; appropriate injection sites include the upper arm, upper thigh, or abdomen.
  • Refrigerate at 2°C–8°C in the original carton; do not freeze.
  • After removal from refrigeration, JUBBONTI® may be stored at room temperature up to 25°C for a maximum of 30 days — cold-chain handling during shipping and storage is a relevant inventory consideration for stocking clinics. [1]

JUBBONTI® Regulatory Status

The FDA approved JUBBONTI® (denosumab-bbdz) in March 2024 as the first biosimilar to PROLIA®, with interchangeable designation, for the full set of PROLIA® indications. [1] JUBBONTI® is a prescription biologic. Regulatory status, pharmacy substitution rules, payer policies, and availability vary by jurisdiction, and practitioners should verify local requirements before clinical use.

FAQs

What is JUBBONTI® and how does it differ from PROLIA®?

JUBBONTI® is denosumab-bbdz, a biosimilar to PROLIA® denosumab sharing the same mechanism, 60mg/mL dose, every-six-month schedule, and approved indications — manufactured by Sandoz rather than Amgen. [1]

Is JUBBONTI® interchangeable with PROLIA®?

Yes. The FDA granted JUBBONTI® interchangeable status in March 2024. Where permitted under applicable state substitution laws, a pharmacist may substitute it for PROLIA® without first obtaining approval from the prescriber.[1]

What is the difference between JUBBONTI® and WYOST®?

JUBBONTI® is the 60mg biosimilar to PROLIA® and is indicated for osteoporosis and other approved bone-loss conditions. WYOST® is the 120mg biosimilar to XGEVA® and is used for approved oncology-related bone indications. The two products differ in dose, reference biologic, and approved uses.

How is JUBBONTI® dosed?

60mg subcutaneously once every 6 months, with adequate calcium and vitamin D supplementation throughout therapy unless clinically contraindicated. [1]

Does JUBBONTI® carry the same Boxed Warning as PROLIA®?

Yes. JUBBONTI® carries a Boxed Warning for severe hypocalcemia in patients with advanced chronic kidney disease, including those receiving dialysis. Existing hypocalcemia should be corrected before treatment begins, and calcium levels should be monitored closely in patients at increased risk.[1]
